CAR-GPC3 T-cells are autologous chimeric antigen receptor T-cell therapies engineered to target glypican-3 (GPC3), a cell-surface glypican highly expressed in hepatocellular carcinoma (HCC) and minimally in healthy adult tissues. These therapies use T cells modified ex vivo to express a synthetic receptor directed at GPC3, conferring tumor antigen specificity and redirecting T-cell cytotoxicity towards GPC3-expressing tumor cells. The mechanism is T-cell mediated cytolysis of target tumor cells following antigen binding. CAR-GPC3 T-cells are under investigation in several clinical trials for advanced HCC and have shown early signals of anti-tumor activity with partial and complete responses observed. Multiple generations exist (including 3rd and 4th generation/armored CARs), some additionally engineered to secrete cytokines or express immune-modulatory signals to enhance efficacy. Clinical development is led by academic groups and companies such as CARsgen Therapeutics, Takeda, and partner institutions[1][4][6].
