Overview

**CAR-iMac** is an engineered macrophage-based cell therapy derived from induced pluripotent stem cells (iPSCs), designed as a next-generation chimeric antigen receptor (CAR) immunotherapy for solid tumors and hematological malignancies. It expresses CARs targeting tumor-associated antigens such as CD19 or mesothelin, enabling antigen-dependent phagocytosis, pro-inflammatory M1 polarization, cytokine secretion (e.g., IL-1β, TNF-α), and tumor microenvironment remodeling to promote T-cell infiltration. Developed through lentiviral transduction, CAR-iMacs demonstrate robust antitumor activity in preclinical models of ovarian cancer, pancreatic cancer, leukemia, and lymphoma, with improved persistence and reduced exhaustion compared to CAR-T cells, positioning it as a promising approach for solid tumor treatment where traditional CAR-T efficacy is limited.
