**CAR-Mix** is an investigational multicellular adoptive cell therapy developed for mesothelin-positive solid tumors. The product combines mesothelin-targeting chimeric antigen receptor macrophages with polyclonal T cells in a single cell-therapy product. The CAR-macrophage component is intended to recognize mesothelin-expressing tumor cells, promote tumor-cell killing and antigen cross-presentation, and activate accompanying polyclonal T cells. Preclinical studies in mesothelin-expressing ovarian cancer models showed enhanced antitumor activity relative to CAR-macrophages or T cells alone, with evidence of T-cell activation and selective expansion of tumor-reactive T-cell clones. An investigator-initiated clinical study has reportedly been initiated.
