CAR NK cell therapy is a first-in-class, off-the-shelf adoptive cell therapy that utilizes natural killer (NK) cells engineered with a chimeric antigen receptor (CAR). Developed by Stephen Strickland and his research group, this therapy is designed to treat relapsed or refractory acute myeloid leukemia (AML). It employs logic-gating technology, enabling the CAR NK cells to identify and target two distinct antigens, CD33 and FLT3, on AML cells, thereby addressing the high heterogeneity of the disease. Additionally, the CAR NK cells incorporate an inhibitory receptor that recognizes healthy cells, preventing off-target effects and minimizing harm to non-cancerous tissues. This approach allows for the development of therapeutic cells from healthy donor cells, which can be stored and made readily available to patients, overcoming the time-consuming and patient-specific limitations of CAR T-cell therapies, especially in cases where patients may have dysfunctional T-cells.
