CAR-NKG2D is an autologous chimeric antigen receptor (CAR) T-cell therapy designed to target NKG2D ligands expressed on cancer cells. Developed by Celyad, following its acquisition of the OnCyte CAR T-cell portfolio from Celdara Medical, which was based on technology originally licensed from Dartmouth College. This therapy involves genetically engineering a patient's T cells to express a CAR based on the natural killer group 2, member D (NKG2D) receptor. The CAR-NKG2D construct typically fuses the human NKG2D receptor with the CD3ζ signaling domain and associates with DAP10 for costimulatory signals, enabling the engineered T cells to recognize and eliminate various solid tumors and hematologic malignancies that express NKG2D ligands. These ligands are stress-induced molecules frequently found on malignant or stressed cells but are typically absent or poorly expressed on healthy tissues.
