Drug intelligence / Profile preview
Car-NTIMP3 (also known as G3C12-NTIMP3) is a synthetic peptide conjugate designed for the treatment of diabetic nephropathy. It consists of the N-terminal domain of human tissue inhibitor of metalloproteinases 3 (TIMP3) conjugated to G3C12, a carrier peptide that selectively targets galectin-3 in the kidney. TIMP3 is an extracellular matrix-bound protein that physiologically inhibits metalloproteinases, particularly A Disintegrin and Metalloproteinase 17 (ADAM17) and matrix metalloproteinases (MMPs) such as MMP2 and MMP9. In diabetic nephropathy, loss of TIMP3 leads to increased ADAM17 and MMP activity, promoting inflammation, oxidative stress, and renal fibrosis. By delivering the active N-terminal domain of TIMP3 specifically to the kidney, Car-NTIMP3 restores TIMP3 inhibitory activity, reduces albuminuria, decreases extracellular matrix deposition, and ameliorates glomerular lesions and podocyte injury.
