CAR-PBMC is an adoptive cell therapy platform developed for the treatment of glioblastoma. Unlike traditional CAR-T therapies that focus on a single immune cell type, this platform utilizes peripheral blood mononuclear cells (PBMCs) to redirect multiple immune cell lineages simultaneously. The cells are engineered using an ultra-rapid flow electroporation process to deliver mRNA encoding multi-targeting chimeric antigen receptors (CARs) and pro-inflammatory cytokines. This approach aims to overcome barriers such as long production times and complex infrastructure requirements by creating a multifunctional immune cell community with a broad range of effector functions against cancer cells. Preclinical studies in orthotopic glioma mouse models and human organotypic brain slice cultures have demonstrated antigen-specific anti-tumor activity and a favorable safety profile, showing no off-target toxicity against neurons, microglia, or astrocytes.
