CAR-PSCA refers to chimeric antigen receptor (CAR) T-cell therapies engineered to target the prostate stem cell antigen (PSCA), a glycosylphosphatidylinositol-anchored cell surface protein overexpressed in several cancers, including prostate, pancreatic, and bladder cancers. In the context of research at Baylor College of Medicine, these CAR-T cells were further modified with a chimeric cytokine receptor (IL4/7 ChR) to overcome the immunosuppressive tumor microenvironment of pancreatic ductal adenocarcinoma (PDAC). This modification allows the cells to utilize inhibitory IL-4 signals to trigger IL-7-mediated proliferative and survival pathways, enhancing persistence and anti-tumor activity.
