CAR-T-19-DNR is an autologous, genetically modified CAR-T cell therapy developed by Immunotech Biopharm (Yongtai Biology). It consists of T cells engineered to express an anti-CD19 chimeric antigen receptor (CAR) along with a dominant-negative mutant type II TGF-β receptor (DNRII). The DNRII component is designed to block TGF-β signaling, which is a common immunosuppressive mechanism in the solid tumor microenvironment. By neutralizing TGF-β's inhibitory effects and preventing the depletion of the CAR-T cells' immune killing ability, the therapy aims to improve the persistence and anti-tumor efficacy of CAR-T cells against CD19-expressing solid tumors, addressing a major limitation of conventional CAR-T therapies in non-hematologic malignancies.
