This is an early-stage preclinical research program investigating chimeric antigen receptor (CAR) T-cell therapies for the treatment of thymic carcinoma. The research focuses on three distinct CAR T-cell constructs targeting trophoblast cell-surface antigen 2 (TROP-2), c-KIT (CD117), and CD70. These targets were selected based on their high surface expression in thymic carcinoma cell lines. The therapy involves engineering T cells via lentiviral transduction to express specific CARs, which then demonstrate antigen-specific cytotoxicity. The project aims to address the lack of effective therapies for this rare mediastinal malignancy and explores multi-antigen strategies, including dual-antigen synNotch CARs, to improve specificity and reduce off-tumor toxicity.
