Overview

This investigational combination immunotherapy, developed by Zhujiang Hospital of Southern Medical University, integrates chimeric antigen receptor (CAR) T-cell therapy with a peptide-specific dendritic cell (DC) vaccine. The therapy is designed for patients with relapsed or refractory acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), and myelodysplastic syndromes (MDS). The DC vaccine component is pulsed with either Eps8 (epidermal growth factor receptor pathway substrate 8) or WT1 (Wilms tumor 1) peptides to induce tumor antigen-specific immune responses. The CAR-T cells are engineered to target a variety of antigens, including CD19, CD20, CD22, and CD10 for ALL, and CD33, CD38, CD56, CD117, CD123, CD34, and MUC1 for AML and MDS. The rationale behind this combination is that the DC vaccine can stimulate and rejuvenate the CAR-T cells in vivo, potentially improving their persistence, expansion, and anti-tumor efficacy compared to CAR-T monotherapy.
