CAR-T cells targeting mesothelin and B7-H3 is an autologous chimeric antigen receptor (CAR) T-cell therapy designed to simultaneously target two tumor-associated antigens: mesothelin (MSLN) and B7-H3 (CD276). Mesothelin is a cell-surface glycoprotein highly expressed in several solid malignancies, including pancreatic and breast cancers, while B7-H3 is an immune checkpoint member of the B7 family that is frequently overexpressed in the tumor microenvironment and linked to immune evasion. By utilizing a dual-targeting approach, this therapy aims to mitigate antigen escape and enhance the precision and potency of T-cell-mediated killing against heterogeneous solid tumors. The Seventh Affiliated Hospital of Sun Yat-sen University is investigating this candidate in early-phase clinical trials for patients with advanced pancreatic cancer and triple-negative breast cancer who have exhausted standard systemic treatment options.
