CAR T cells with thermal switch is a research-stage platform for remotely controlled immunotherapy developed by researchers at the Georgia Institute of Technology and Emory University. The technology utilizes genetically engineered T cells containing synthetic thermal gene switches with Heat Shock Elements (HSEs). These switches trigger the expression of therapeutic transgenes—such as chimeric antigen receptors (CARs), bispecific T cell engagers (BiTEs/TCEs), or cytokines like IL-15—only when exposed to mild hyperthermia (40-42°C). Heat is delivered noninvasively to the tumor site using magnetic resonance imaging-guided focused ultrasound (MRgFUS) or photothermal methods. This spatiotemporal control aims to enhance safety by limiting systemic toxicity and overcome tumor heterogeneity by inducing local production of T-cell engagers.
