CAR-T-EGFR is a chimeric antigen receptor T cell therapy engineered to target the epidermal growth factor receptor (EGFR) expressed on tumor cells. This cell therapy consists of autologous or allogeneic T cells genetically modified to express a CAR specific for EGFR. The intervention aims to harness immune-mediated cytotoxicity against EGFR-expressing solid tumors, such as glioblastoma or EGFR-mutant non-small cell lung cancer (NSCLC). Preclinical and early clinical data suggest that while CAR-T-EGFR can cause tumor regression, especially in glioblastoma and drug-tolerant/resistant NSCLC cell populations, challenges remain in efficacy due to the immunosuppressive tumor microenvironment and risk of on-target, off-tumor toxicity given the expression profile of EGFR. EGFR CAR-T therapies may be used alone or in combination with other agents, such as tyrosine kinase inhibitors (TKIs) or TGF-β pathway modulators, to enhance antitumor activity[1][3][5].
