CAR T-REX (CAR T Cells Rewired to Prevent EXhaustion in the Tumour Microenvironment) is a research program and European consortium project led by Stemmatters. The initiative aims to overcome the limitations of CAR T-cell therapy in solid tumors by engineering T cells that are resistant to exhaustion within the immunosuppressive tumor microenvironment (TME). The technology utilizes non-viral gene delivery and precise genome editing to create auto-regulated genetic circuits controlled by microRNAs. These circuits modulate T-cell autoregulatory pathways to maintain anti-tumor activity and persistence. The project's lead candidates target HER2/Neu-positive cancers, specifically breast and gastric carcinomas, and are currently in preclinical development as part of a four-year research initiative funded by the European Innovation Council.
