CAR T targeting CD123 refers to a class of chimeric antigen receptor (CAR) T-cell therapies engineered to recognize and eliminate cells expressing CD123 (interleukin-3 receptor alpha chain). CD123 is a cell surface protein frequently overexpressed in various hematological malignancies, including acute myeloid leukemia (AML), blastic plasmacytoid dendritic cell neoplasm (BPDCN), and certain types of lymphoid leukemia, while remaining low or absent on normal hematopoietic stem cells. These therapies involve the genetic modification of autologous or allogeneic T cells to express a CAR construct that redirects T-cell-mediated cytotoxicity toward CD123-positive blasts. Several investigational products are in clinical development, such as Mustang Bio's MB-102 and Cellectis's UCART123, targeting indications with high unmet medical needs.
