car.b7-h3t cells are a preparation of autologous T lymphocytes that have been genetically engineered to express a chimeric antigen receptor (CAR) targeting B7-H3 (CD276), an immunoregulatory ligand in the B7 family. The CAR construct typically includes a single-chain variable fragment (scFv) derived from an anti-B7-H3 antibody, fused with intracellular signaling domains such as CD28, 4-1BB, and CD3ζ. These engineered T cells are designed to recognize and kill tumor cells expressing B7-H3, which is highly expressed in many solid tumors—including osteosarcoma, neuroblastoma, glioma, breast cancer, prostate cancer, pancreatic cancer—and has limited expression in normal tissues. The therapy aims to overcome immune evasion by tumors and is being investigated primarily for relapsed or refractory solid malignancies[1][2][5].
