Drug intelligence / Profile preview
CAR.CD123 NK cells is an investigational, off-the-shelf, second-generation chimeric antigen receptor (CAR) natural killer (NK) cell therapy targeting CD123 (interleukin-3 receptor subunit alpha). Developed by the Bambino Gesù Children's Hospital, the therapy utilizes healthy donor-derived peripheral blood NK cells engineered with a retroviral vector expressing an anti-CD123 CAR construct containing a 4-1BB costimulatory domain and a CD3ζ signaling domain. It is designed as an allogeneic cellular immunotherapy for the treatment of CD123-positive hematological malignancies, specifically pediatric acute myeloid leukemia (AML), offering a potentially safer profile with reduced risk of graft-versus-host disease (GvHD) and cytokine release syndrome (CRS) compared to conventional CAR-T cell therapies.
