Overview

CAR.HER2.TR2.41BB T cells are genetically engineered chimeric antigen receptor (CAR) T cells designed to target HER2-expressing solid tumors, such as breast cancer, while simultaneously overcoming the immunosuppressive tumor microenvironment (TME). Developed by researchers at the Baylor College of Medicine, these T cells co-express a second-generation CAR targeting human epidermal growth factor receptor 2 (HER2) and a novel chimeric costimulatory receptor (TR2.41BB). The TR2.41BB receptor consists of a single-chain variable fragment (scFv) derived from the TRAIL receptor 2 (TR2) agonist antibody DS-8273a fused to a 4-1BB (CD137) costimulatory endodomain. This dual-targeting approach allows the T cells to eliminate HER2-positive tumor cells while selectively binding to TR2 expressed on myeloid-derived suppressor cells (MDSCs), inducing MDSC apoptosis and delivering a costimulatory signal that enhances T-cell survival, proliferation, and persistence at the tumor site.
