CAR.MUC1.TR2.41BB T cells is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to treat solid tumors, specifically breast cancer. Developed by researchers at the Baylor College of Medicine, these T cells are genetically engineered to express a CAR targeting tumor-associated mucin 1 (MUC1) alongside a novel chimeric costimulatory receptor (TR2.41BB) that targets tumor necrosis factor-related apoptosis-inducing ligand receptor 2 (TR2 / TNFRSF10B) expressed on myeloid-derived suppressor cells (MDSCs). By binding to TR2, the costimulatory receptor induces apoptosis in immunosuppressive MDSCs, thereby remodeling the tumor microenvironment and restoring the cytotoxic activity of the CAR T cells. Simultaneously, this interaction triggers intracellular 4-1BB (CD137) costimulatory signaling within the T cells, enhancing their proliferation, persistence, and anti-tumor efficacy.
