Overview

CAR19-CYNK is an allogeneic, off-the-shelf **CAR-NK cell therapy** developed by Celularity for CD19-positive B-cell malignancies, including B-cell lymphomas. The product consists of human placental CD34-positive hematopoietic stem cell-derived natural killer cells that are retrovirally transduced to express an anti-CD19 chimeric antigen receptor incorporating a CD19 single-chain variable fragment with CD28 and CD3 zeta signaling domains. After expansion in cytokine-containing culture, the cells are formulated as a cryopreserved biologic intended for adoptive cellular immunotherapy. Its mechanism is to direct NK-cell cytotoxicity toward CD19-expressing malignant B cells, with preclinical studies showing enhanced in vitro killing and survival benefit in lymphoma xenograft models.
