Overview

CAR19-TIM-3-Fc is an engineered, bicistronic chimeric antigen receptor T cell therapy that combines CD19-directed CAR T cells with a secreted TIM-3-Fc decoy receptor. The construct is designed to address immune escape and poor persistence seen with conventional CD19 CAR-T in relapsed/refractory B-cell acute lymphoblastic leukemia (B-ALL). By secreting a TIM-3-Fc decoy, the therapy blocks the TIM-3/galectin-9 immune checkpoint axis, overcoming the immunosuppressive bone marrow microenvironment, enhancing antileukemic activity, and promoting long-term persistence and expansion of effector/memory T cells in vivo. Preclinical studies demonstrate improved outcomes in B-ALL models, indicating promise for refractory patients.[1][2][6][7]
