Overview

CAR19-tTreg is an investigational cell therapy consisting of regulatory T cells (Tregs) that have been genetically engineered to express a chimeric antigen receptor (CAR) targeting CD19, a protein commonly found on B cells. Unlike conventional CAR-T therapies, which use effector T cells to kill target cells, CAR19-tTreg uses Tregs to suppress immune responses in a targeted manner. The goal is to induce antigen-specific immunosuppression for conditions such as graft-versus-host disease (GvHD), autoimmune diseases, and organ transplantation. By redirecting the suppressive function of Tregs specifically toward CD19-expressing cells or tissues, this approach aims to promote immune tolerance while minimizing systemic immunosuppression. Preclinical studies have shown that CAR-engineered Tregs maintain their regulatory phenotype and can home to target tissues with enhanced suppressive efficacy compared to polyclonal Tregs[8]. Clinical trials are ongoing or planned for applications such as prevention of transplant rejection and treatment of autoimmune disorders.
