Overview

CAR19T2 is an experimental third-generation autologous Chimeric Antigen Receptor (CAR) T-cell therapy designed to treat B-cell hematological malignancies. It features a humanized single-chain variable fragment (scFv) specific for the **B-lymphocyte antigen CD19**. A defining characteristic of CAR19T2 is its unique costimulatory architecture, which incorporates both the **CD28** domain and a **Toll-like receptor 2 (TLR2)** domain, alongside the standard **CD3 zeta** signaling chain. The inclusion of the TLR2 costimulatory domain is intended to mimic innate immune signaling, potentially enhancing the activation, metabolic fitness, and persistence of the CAR-T cells within the immunosuppressive tumor microenvironment. It has been primarily investigated for the treatment of refractory or relapsed B-cell acute lymphoblastic leukemia (B-ALL) and B-cell lymphoma, particularly in pediatric patients.
