Overview

**CAR2019(AS)** is an investigational autologous tandem CD19/CD20 chimeric antigen receptor T-cell therapy for relapsed or refractory B-cell lymphoma. It is an optimized version of the earlier CAR2019 construct, incorporating protease cleavage sites between the antigen-binding single-chain variable fragment regions and hinge regions to improve CAR function, persistence, and resistance to T-cell exhaustion. The engineered T cells recognize both CD19 and CD20 on malignant B cells and mediate targeted cytotoxic killing. In a multicenter phase 1 study in heavily pretreated relapsed or refractory B-cell lymphoma, CAR2019(AS) produced high response rates with manageable cytokine-release syndrome and no reported grade 3 or higher neurotoxicity.
