CAR72_21 NK cells is an investigational, allogeneic "off-the-shelf" cell therapy consisting of cord blood-derived natural killer (NK) cells engineered to express a chimeric antigen receptor (CAR) targeting CD72. The CAR construct is nanobody-based (VHH-derived) and incorporates a 4-1BB costimulatory domain. To enhance therapeutic efficacy and persistence in the immunosuppressive environment of B-cell malignancies, the cells are "armored" to co-express interleukin-21 (IL-21). Developed for the treatment of chronic lymphocytic leukemia (CLL) and Richter transformation (RT), CAR72_21 has demonstrated potent in vitro cytotoxicity against tumor cell lines and primary patient samples, showing potential synergy when combined with antibodies like obinutuzumab.
