Drug intelligence / Profile preview
Carabrone is a natural sesquiterpene lactone isolated from the traditional Chinese herb *Carpesium abrotanoides* L. and other *Carpesium* species. It is being investigated as a preclinical therapeutic candidate for various inflammatory, metabolic, and neurodegenerative disorders. Carabrone acts as a multi-target inhibitor, directly binding to the NACHT domain of the NLR family pyrin domain-containing 3 (NLRP3) inflammasome to prevent its activation and assembly. Additionally, it directly binds to and inhibits signal transducer and activator of transcription 3 (STAT3) activation, and covalently targets ubiquitin-conjugating enzyme E2 D3 (UBE2D3) at the cysteine 85 residue to suppress NF-κB signaling. Through these mechanisms, carabrone has demonstrated therapeutic efficacy in animal models of Alzheimer's disease, multiple sclerosis, metabolic dysfunction-associated steatohepatitis (MASH), gouty arthritis, sepsis, and pancreatic cancer.
