Carba-nicotinamide adenine dinucleotide (carba-NAD) is a synthetic analog of nicotinamide adenine dinucleotide (NAD+), designed to mimic the structure and function of NAD+ but with improved hydrolytic stability. The modification involves replacing the ribose ring in the nicotinamide moiety with a carbocyclic ring. This makes carba-NAD more resistant to enzymatic hydrolysis by NAD+-consuming enzymes such as adenosine diphosphate ribosyl transferases and NAD glycohydrolases. It acts as a substrate for some NAD+-handling enzymes like alcohol dehydrogenases and can inhibit certain ART enzymes and sirtuins (e.g., Sirt3/5). Carba-NAD is primarily used as a research tool in biochemical studies—especially for protein crystallization or enzyme assays requiring stable NAD+ mimics—and is not approved for therapeutic use[9][2][10].
