Carbazole Blue (CB) is a synthetic small molecule analogue of carbazole containing a triphenylmethane pharmacophore. Originally synthesized as a potential NADPH oxidase (NOX) inhibitor, Carbazole Blue has been shown to directly bind to DNA and inhibit cancer growth and metastasis. It selectively targets DNA-related processes, such as aberrant DNA replication and DNA repair machinery, which tumor cells rely on for survival, thereby inducing G1/S cell cycle arrest and apoptosis. In preclinical models of triple-negative breast cancer (TNBC) and estrogen receptor (ER)-positive/mutant breast cancers, systemic delivery of Carbazole Blue (including via nanoparticle-based approaches) significantly suppressed tumor growth and metastasis without inducing systemic toxicity.
