This is a combination therapy that includes carboplatin, chloroquine (or hydroxychloroquine), and gemcitabine. The combination works by targeting cancer cells through multiple mechanisms. Carboplatin and gemcitabine are chemotherapy drugs that destroy quickly dividing cells, while chloroquine/hydroxychloroquine inhibits late-phase autophagy. Autophagy is a catabolic process triggered in cells during periods of metabolic or hypoxic stress that enables their survival. By inhibiting late-stage autophagy, chloroquine/hydroxychloroquine enhances cell death due to accumulation of damaged organelles, potentially making cancer cells more susceptible to the cytotoxic effects of chemotherapy.
