Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents—carboplatin, cyclophosphamide, and doxorubicin. Each drug has a distinct mechanism of action targeting cancer cells at different stages of the cell cycle: - **Carboplatin** is a platinum-based compound that forms DNA crosslinks, inhibiting DNA synthesis and function, leading to apoptosis. - **Cyclophosphamide** is an alkylating agent that crosslinks DNA strands and decreases DNA synthesis, particularly effective during the resting phase (G0) of the cell cycle. - **Doxorubicin** is an anthracycline antibiotic that intercalates into DNA and inhibits topoisomerase II, preventing replication and transcription at any point in the cell cycle. This combination has been studied as first-line therapy for advanced epithelial ovarian carcinoma (stage III-IV), where it demonstrated similar efficacy to cisplatin-based regimens in terms of response rate, progression-free survival (PFS), and overall survival. The regimen may also be considered for other solid tumors where these agents are individually active[1].
