Overview

This combination has been studied primarily in the context of advanced or refractory cancers such as pancreatic cancer and clear cell carcinoma. Itraconazole is included for its ability to overcome drug resistance mechanisms—particularly by inhibiting P-glycoprotein-mediated efflux—and to enhance the efficacy of cytotoxic agents. Clinical studies have shown promising response rates and overall survival benefits in second-line settings for pancreatic cancer, with acceptable toxicity profiles[1][2][3][4]. The regimen is typically administered in cycles every two weeks.
