Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents—carboplatin, docetaxel, and irinotecan. Each drug has a distinct mechanism of action: - Carboplatin is an organoplatinum alkylating agent that forms DNA crosslinks and inhibits DNA synthesis and function. - Docetaxel is a taxane derivative that stabilizes microtubules and prevents their depolymerization, thereby inhibiting cell division. - Irinotecan is a topoisomerase I inhibitor that prevents the religation of single-strand breaks in DNA during replication. The combination has been studied in various solid tumors including carcinoma of unknown primary (CUP) and other advanced cancers. It aims to exploit the synergistic effects of these agents with non-overlapping toxicities to improve antitumor efficacy[1][2]. This regimen may be used in clinical trials or for patients with good performance status who can tolerate intensive chemotherapy.
