Overview

Carboplatin + doxorubicin is a combination chemotherapy regimen consisting of two cytotoxic agents used primarily in the treatment of ovarian cancer and other solid tumors. Carboplatin is a platinum-based compound that induces DNA crosslinking and apoptosis by forming covalent bonds with DNA bases, thereby inhibiting DNA replication and transcription. Doxorubicin is an anthracycline antibiotic that intercalates into DNA, inhibits topoisomerase II activity (leading to double-strand breaks), and generates free radicals causing further cellular damage. The combination—often using pegylated liposomal doxorubicin (PLD) for improved tolerability—has demonstrated efficacy in recurrent or relapsed ovarian cancer, particularly in patients who are platinum-sensitive or have progressed on single-agent therapy. This regimen aims to maximize antitumor activity while reducing overlapping toxicities compared to other combinations such as carboplatin with paclitaxel[1][2][3][4].
