Overview

This is a combination chemotherapy regimen consisting of four cytotoxic agents: carboplatin, fluorouracil (5-FU), lomustine, and vinorelbine. Each drug has a distinct mechanism of action targeting cancer cells through different pathways: - **Carboplatin** is a platinum-based compound that forms DNA crosslinks, inhibiting DNA synthesis and function. - **Fluorouracil** is an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis. - **Lomustine** is an alkylating nitrosourea agent that causes DNA and RNA crosslinking and strand breaks. - **Vinorelbine** is a vinca alkaloid that disrupts microtubule formation during mitosis. This combination has been studied for the treatment of brain metastases from breast or lung cancer. The regimen aims to maximize anti-tumor efficacy by combining drugs with complementary mechanisms while managing toxicity profiles. Clinical studies have shown partial remissions in patients with brain metastases using this schedule[1].
