This is a combination chemotherapy regimen consisting of three cytotoxic agents—carboplatin, gemcitabine, and paclitaxel. Each drug has a distinct mechanism of action targeting cancer cell proliferation: - **Carboplatin** is a platinum-based compound that forms DNA crosslinks, inhibiting DNA synthesis and function. - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase. - **Paclitaxel** stabilizes microtubules, preventing their depolymerization during cell division. The combination has been studied primarily in advanced urothelial carcinoma and other solid tumors. Clinical trials have shown this regimen to be active with manageable toxicity profiles in patients with advanced urothelial malignancies[1][2][3]. The most common toxicities include neutropenia, thrombocytopenia, anemia, neuropathy, and gastrointestinal symptoms. This regimen may be used when cisplatin-based therapy is not suitable or as an alternative for certain patient populations.
