This is a combination chemotherapy regimen consisting of three anticancer drugs: carboplatin (a platinum-based agent), gemcitabine (a nucleoside analog), and pemetrexed (a multitargeted antifolate). The combination has been studied in various configurations for the treatment of non-small cell lung cancer (NSCLC). Pemetrexed inhibits three enzymes in the folate pathway involved in nucleotide synthesis: thymidylate synthase, dihydrofolate reductase, and glycinamide ribonucleotide formyl transferase. Carboplatin forms crosslinks with DNA, interfering with cell division. Gemcitabine is a nucleoside analog that replaces cytidine during DNA replication, causing cell death.
