Overview

The combination of carboplatin, ifosfamide, and paclitaxel (often referred to as the TICb regimen) is a multi-agent chemotherapy protocol primarily investigated for the treatment of advanced or recurrent malignant mixed Müllerian tumors (MMMTs) of the uterus and adnexa. This regimen leverages three distinct mechanisms: - Paclitaxel is a microtubule-stabilizing agent that inhibits cell division. - Ifosfamide is an alkylating agent that induces DNA cross-linking and damage. - Carboplatin is a platinum-based compound that also causes DNA cross-linking. Preclinical data suggest synergistic effects when these agents are combined, particularly with paclitaxel administered prior to alkylating agents. Clinical studies have shown this combination yields favorable response rates and manageable toxicity in patients with advanced gynecologic cancers[1][3][6]. The regimen has also been explored in other solid tumors such as germ cell tumors[2].
