This is an experimental combination therapy consisting of three agents: - **Carboplatin** is a platinum-based chemotherapy that causes DNA crosslinking and apoptosis in rapidly dividing cells. - **Inupadenant** (EOS100850) is a small molecule antagonist of the adenosine A2A receptor, designed to counteract immunosuppression within the tumor microenvironment and enhance anti-tumor immune responses. - **Pemetrexed** (Alimta) is an antifolate antineoplastic agent that inhibits multiple enzymes involved in folate metabolism and DNA synthesis, leading to inhibition of cancer cell growth. The combination has been studied primarily for metastatic non-squamous non-small cell lung cancer (NSCLC) after progression on prior immunotherapy. In Phase 2 clinical trials, this regimen demonstrated manageable safety and tolerability with no dose-dependent toxicities. The overall response rate was reported at 63.9%, with a median progression-free survival of 7.7 months across all patients[1][2][9]. Inupadenant’s mechanism aims to restore immune activity suppressed by adenosine signaling following chemotherapy[1][2].
