This is a combination regimen consisting of three agents: - **Carboplatin** (a platinum-based chemotherapeutic that induces DNA crosslinking and damage, leading to apoptosis in rapidly dividing cells), - **Ipatasertib** (an orally administered, highly selective pan-AKT inhibitor that blocks the PI3K/AKT signaling pathway involved in cell proliferation and survival), and - **Paclitaxel** (a taxane-class chemotherapeutic that stabilizes microtubules, inhibiting cell division). The combination is under investigation for use as neoadjuvant or metastatic therapy in cancers such as triple-negative breast cancer (TNBC) and epithelial ovarian cancer. The rationale for this regimen is to combine cytotoxic chemotherapy with targeted inhibition of the AKT pathway, which may enhance antitumor activity especially in tumors harboring PI3K/AKT/PTEN alterations. Early-phase clinical trials have shown the combination to be safe with modest efficacy[1][5][6][8].
