Carboplatin + leronlimab is an investigational combination therapy being studied primarily for the treatment of CCR5-positive metastatic triple-negative breast cancer (mTNBC). Carboplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis in rapidly dividing cells. Leronlimab (PRO 140) is a humanized IgG4 monoclonal antibody that acts as an antagonist of the C-C chemokine receptor type 5 (CCR5), which plays roles in tumor metastasis, immune cell trafficking, and HIV infection. The combination aims to leverage the cytotoxic effects of carboplatin with the immunomodulatory and anti-metastatic properties of CCR5 blockade by leronlimab. Early-phase clinical trials have shown promising results including decreased cancer-associated macrophage-like cells and improved progression-free survival in mTNBC patients[1][2][3][4][7][9]. Leronlimab has received FDA fast track designation for use with carboplatin in this setting[5].
