A combination therapy consisting of carboplatin, metformin, and pemetrexed. This regimen combines standard platinum-based chemotherapy (carboplatin and pemetrexed) with metformin, an antidiabetic drug that has shown anticancer properties. The combination has been studied primarily for non-small cell lung cancer (NSCLC), with particular interest in STK11-mutated NSCLC, a subgroup inherently resistant to immune checkpoint inhibitors. Metformin appears to enhance the effectiveness of chemotherapy by decreasing PKM2 expression, inhibiting glucose metabolism in cancer cells, and demonstrating antiproliferative and antiangiogenic effects.
