Overview

This is a combination regimen consisting of **carboplatin** (a platinum-based DNA-damaging chemotherapeutic), **paclitaxel** (a microtubule-stabilizing chemotherapeutic), **cobolimab** (an investigational monoclonal antibody against TIM-3), and **dostarlimab** (a monoclonal antibody checkpoint inhibitor targeting PD-1). This combination is under investigation or in clinical trials for various advanced solid tumors, including gynecological malignancies and possibly others. - **Carboplatin** forms DNA adducts, triggering apoptosis in rapidly dividing cells (antineoplastic, cytotoxic). - **Paclitaxel** inhibits microtubule disassembly, preventing mitosis (antineoplastic, cytotoxic). - **Cobolimab** is an investigational anti-TIM-3 immune checkpoint inhibitor, aiming to restore interferon signaling and enhance cytotoxic T cell activity. - **Dostarlimab** (marketed as Jemperli) is an FDA-approved anti-PD-1 antibody used in certain endometrial cancers and under study for other tumors; it blocks the PD-1 receptor, releasing immune inhibition and promoting antitumor T cell responses[2][4][8]. Combining both established chemotherapy (carboplatin, paclitaxel) and dual immune checkpoint inhibition (TIM-3 and PD-1) is intended to enhance anti-tumor efficacy by both killing tumor cells directly and augmenting the immune system’s ability to control cancer.
