Overview

This combination regimen consists of the platinum-based chemotherapeutic agent carboplatin, the taxane microtubule stabilizer paclitaxel, and the anti-PD-1 monoclonal antibody pembrolizumab. It is being investigated by Brown University as a reduced-intensity induction therapy for frail geriatric patients (aged 65 and older) with stage IB-IVA squamous cell carcinoma of the head and neck (SCCHN). The regimen aims to provide a tolerable yet effective treatment option for patients who may not withstand standard high-intensity cisplatin-based chemoradiation. Carboplatin and paclitaxel provide cytotoxic debulking, while pembrolizumab leverages the immune system by blocking the PD-1/PD-L1 checkpoint, potentially enhancing the long-term response and synergy with subsequent radiation therapy.
