Overview

This is a combination regimen consisting of three agents: - **Carboplatin** is a platinum-based chemotherapy that causes DNA crosslinking and inhibits DNA synthesis, leading to apoptosis in rapidly dividing cancer cells. - **Paclitaxel** is a taxane-class chemotherapeutic that stabilizes microtubules and prevents their depolymerization, thereby inhibiting cell division. - **Penpulimab** (also known as Anike or Penpulimab-kcqx) is an engineered humanized IgG1 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. By blocking PD-1, penpulimab enhances T-cell mediated anti-tumor immune responses. Unlike other PD-1 antibodies, penpulimab’s Fc region has been modified to reduce immune-related adverse events by minimizing Fcγ receptor binding and complement activation. This triple combination represents an integration of cytotoxic chemotherapy with immunotherapy. It has been investigated primarily for advanced solid tumors including nasopharyngeal carcinoma (NPC), where it may be used as first-line therapy in combination with platinum-based chemotherapy regimens[8][10]. Each component has established use in oncology; the addition of penpulimab aims to improve outcomes by leveraging both direct tumor cytotoxicity and immune-mediated effects.
