This is a combination therapy consisting of three agents: - **Carboplatin** is a platinum-based chemotherapeutic agent that induces DNA crosslinking, leading to apoptosis in rapidly dividing cells. - **Paclitaxel** is a microtubule-stabilizing agent that inhibits cell division by preventing microtubule depolymerization. - **Tigatuzumab** (CS-1008) is a humanized monoclonal antibody targeting death receptor 5 (DR5, also known as TNFRSF10B or TRAIL-R2), which induces apoptosis selectively in tumor cells overexpressing DR5. Tigatuzumab acts as an agonist of DR5 and has been investigated for its antitumor activity in various solid tumors. The combination has been studied primarily for the treatment of chemotherapy-naïve patients with metastatic or unresectable non-small cell lung cancer (NSCLC). In clinical trials, this regimen was well tolerated but did not significantly improve efficacy compared to carboplatin/paclitaxel alone in unselected advanced NSCLC patients[1][4][6].
