Overview
This is a combination chemotherapy regimen consisting of three agents: - **Carboplatin** is a platinum-based chemotherapeutic that forms DNA crosslinks, inhibiting DNA synthesis and function. - **Pegylated liposomal doxorubicin (PLD)** (marketed as Caelyx or Doxil) is an anthracycline antibiotic encapsulated in liposomes with polyethylene glycol to improve pharmacokinetics and reduce toxicity. It intercalates into DNA and inhibits topoisomerase II, leading to apoptosis. - **Doxorubicin** is also an anthracycline antibiotic with the same mechanism as PLD but without the liposomal formulation. The combination of carboplatin and PLD has demonstrated efficacy in recurrent ovarian cancer, particularly in platinum-sensitive disease. The addition of standard (non-liposomal) doxorubicin to this regimen is not standard practice but may be considered in certain clinical scenarios for enhanced cytotoxic effect. The primary indication for this combination is recurrent epithelial ovarian carcinoma[1][2][3].
