Arm B of the Fox Chase Cancer Center Phase II trial (NCT03786692) represents a combination chemotherapy and anti-angiogenic regimen consisting of carboplatin, pemetrexed, and bevacizumab. This triplet serves as the control arm in a study evaluating the addition of the PD-L1 inhibitor atezolizumab for patients with Stage IV non-squamous non-small cell lung cancer (NSCLC) who either harbor sensitizing EGFR mutations (following progression on tyrosine kinase inhibitors) or are never smokers with wild-type tumors. Carboplatin is a platinum-based cytotoxic agent that causes DNA cross-linking; pemetrexed is an antifolate that disrupts metabolic processes dependent on folate by inhibiting multiple enzymes; and bevacizumab is a monoclonal antibody that inhibits angiogenesis by targeting vascular endothelial growth factor (VEGF). This combination is a standard-of-care alternative to paclitaxel-based regimens in non-squamous NSCLC.
