Drug intelligence / Profile preview
This combination consists of four agents: - **Carboplatin:** a DNA-damaging platinum-based chemotherapeutic that causes inter- and intra-strand DNA crosslinking, leading to apoptosis in rapidly proliferating cells. - **Pemetrexed:** an antifolate antineoplastic that inhibits multiple folate-dependent enzymes involved in purine and pyrimidine synthesis, arresting DNA and RNA synthesis. - **Cobolimab:** a monoclonal antibody targeting T cell immunoglobulin and mucin domain-containing protein 3 (TIM-3), functioning as an immune checkpoint inhibitor to promote T cell activation and antitumor immune response[2][4][6]. - **Dostarlimab:** a monoclonal antibody targeting programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1 and PD-L2, preventing T cell inhibition and promoting antitumor immunity[2][4][6]. This combination brings together cytotoxic chemotherapy (carboplatin, pemetrexed) and dual immune checkpoint blockade (TIM-3 and PD-1 inhibition) to address tumor cell burden and modulate the immune microenvironment for enhanced antitumor effects. This combination has been explored in clinical trials primarily for high-risk solid tumors (notably melanoma and advanced NSCLC in various related regimens), with recent interest in the neoadjuvant and perioperative setting in melanoma[2][4][6].
