Overview

Carboplatin + thymidine is an experimental combination therapy consisting of the platinum-based alkylating agent carboplatin and the nucleoside thymidine. Carboplatin exerts its antineoplastic effects by forming DNA crosslinks, thereby inhibiting DNA synthesis and function, leading to apoptosis in rapidly dividing cells. Thymidine, a naturally occurring nucleoside, has been investigated for its potential to modulate chemotherapy toxicity and enhance efficacy. In phase I clinical studies, intravenous thymidine was administered as a 24-hour infusion prior to carboplatin dosing in patients with refractory cancers. The combination aimed to evaluate whether thymidine could protect against certain toxicities of carboplatin (notably thrombocytopenia) without diminishing anticancer activity. Early results indicated that the regimen was well tolerated and showed some evidence of clinical benefit in high-grade glioma and metastatic colon cancer[1].
